This is not misandry, in just the same way as having a woman antagonist in a story is not misogyny. There is no direct or implied generalization here, it just shows a single guy being an asshole. If you feel attacked by this comic, it’s time for some serious introspection.
This comic on its own is not misandry, but Pizzacake sure is a mysandrist due to the frequency at which they depict male characters as the badguys in their strips. It’s certainly quite notable.
As a guy I don’t see how it affects me. It’s literally one man shown in the image. I did not get a feeling of “Great now they’ll think we’re all like that” or “Oh geez you mean people hate when I say those things to them?”. All I have to do to not be lumped in with that guy is to not do that one specific thing in the comic.
Or it’s just turning very common and relatable occurrences of the lesbian and trans experiences into a comic?

I’m avoiding going into too much detail precisely because that’s what the comment is meant to trigger. If he had enough gumption to come out and say what he wanted to say and get the comment removed that’d be one thing. I think they’re trying to voice their support of the guy in this comic by stating that, “They’re just telling them what they’ve got to offer. that isn’t a problem”.
They’re a mod on a conservative community with a ton of alts on different instances each with significant numbers of entries in the modlogs, I’d say they can be ignored.
